Moreover, you will notice the various ultrastructural changes
during apoptotic process, if you use several different cultured
cells or tissues, and even' if using the same cell line treated
with different stimuli such as heat shock and anti-cancer drugs
(Fig. 8).
The conventional ISEL technique stains not only apoptotic
cells but necrotic cells. The ISELITEM technique clearly demonstrates many immunogold particles indicating the sites of 3' -OH
ends in the nuclei of necrotic cells (Fig. 9) and supports the above
finding of the conventional ISEL technique.
When you use image analysis combined with the ISELITEM
technique, you can semi-quantify DNA strand breaks in cells undergoing apoptosis and necrosis. The image analysis for quan-
