The formation, size, and stability of A8-35
particles and MP/A8-35
complexes were studied in the presence or absence of detergent or
excess APols, providing insights into the rate of exchange of
surfactants between particles.
In the course of a study of bacteriorhodopsin (BR)/A8-35 complexes,
further data were collected on the behavior of A8-35
particles. The
techniques listed are those relevant to these particles.
First-generation glucose-based NAPols were obtained by
co-telomerization of hydrophilic and amphipathic monomers.
Glucosylated NAPols similar to those in Study 4.13 were obtained by
grafting hydrophobic chains onto a hydrophilic homopolymer.
The size of SMA/DMPC complexes (SMALPs) was studied by DLS
and EM after negative staining and their organization by solution
NMR and EPR. The structure of SMA is given in Fig. 4.1e and a
scheme of the SMALP particles in Fig. 4.27a.
The critical aggregation concentration (CAC) of A8-35
was
determined both by tensiometry and by FRET using two
complementary fluorescent derivatives of A8-35.
Homopolymeric glucose-based NAPols were obtained by
telomerization of an amphipathic precursor. Their structure is shown
in Fig. 4.5d.
The structure and dynamics of A8-35
particles were studied by
all-atom and coarse-grained molecular dynamics simulations.
