[14]. The evanescent wave exponentially decays with a characteristic decay length of
~100 nm [15]. Hence, the evanescent wave illuminates the thin region relatively
close to the surface, as compared with the epi-illumination, where the light goes
deeply into the water phase (Panel b). As a result, the number of fluorophores excited
