The Iatroscan TLC/FID is a good and relevant technique for rhe study of
marine lipids as biological tracers. Although applications are numerous
in the literature, in many cases, it appears to be difficult to obtain a
good separation between several classes of lipids, which contribute to
the degradation index. Generally, when peak resolution is effective,
standard deviation does not exceed 8% (n = 3), but the low resolution
of sterols and diacylglycerols peak in a first case and of pigments and
monoacylglycerols peaks in a second case give highest standard deviations (up to l6%, n=3). A new Chromarod thin-layer chromatography
procedure was developed for improving the separation of lipid degradation products in neutral and polar fractions. It was thus possible to
separate 1,2 and 1,3 diacylglycerols isomers from sterols with a high
repeatability. Monoacylglycerols were co-eluted with yellow pigments
but well separated from chlorophyll a, monogalactosyldiacylglycerols
and digalactosyldiacylglycerols peaks. The rotal development time was
168 minutes. This procedure was successfully applied to rhe study of
lipid classes in sinking particles.
