contour line will affect the karyotyping results. Careful drawing and
editing of the contour lines is necessary.
Results
Metaphase spread analysis
Complex rearrangements in tumor cells are mapped rapidly by SKY using
tumor metaphase cell preparations. Figure 1demonstrates this by showing
the SKY analysis of a metaphase spread prepared from the human prostate
cancer cell line TSUPRI. Here, the DAPI image (Fig. lA) helped to identify
chromosomes. Following chromosome classification, individual chromosomes were assigned 'classification colors' which linked the fluorescence
spectra to chromosome-specific probe mixtures (Fig. IB, C). SKY analysis
of TSUPRI metaphases revealed a number of previously unknown translocations (t(2;8),t(3;19),t(6;7),t(6;15),t(15;18» in the presence of several
marker chromosomes (Fig. ID). In summary, Fig. lA shows the inverted
image of the chromosome spread acquired through the DAPI filter. Figure
IB shows the same chromosomes in an RGB pseudo-color image acquired
through the SKY filter. Based on the measurement of the complete spectrum for each point in this metaphase image, a spectral classification algorithm was applied that allowed the assignment of a pseudo-color to all
points in the image that had the same spectrum. A karyotype table was
obtained as shown in Fig. 1C. There were 84 chromosomes in this spread,
with six of them classified as marker chromosomes. We changed the format of this karyotype table as shown in Fig. ID, demonstrating chromosomes in classification colors to the left and normal idiograms to the right.
Clearly, translocations in the cell of this prostate cancer cell line involved
the chromosomes 3, 7, 8, 15, 18, 19.
Chromosome enumeration in interphase cells
Spectral Imaging can be applied to routinely score eight or more chromosomes in single interphase cell nuclei. The following examples show SIm
results using three different cell types hybridized with a 8-chromosome
