Cells and cell masses come out from the cut opening (Fig. 3). They attach to the bottom of the culture vessels, and make foci of proliferating
cells. In the meantime, larval fragments may continue to exhibit twitching movements. Most mosquito cell lines have been established using
this method (e.g., Singh, 1967; Mitsuhashi, 1981) .
• • Comments
If larvae that have been damaged only slightly are involved in the culture, they swim around in the medium, and may eat migrated cells. Be
sure that such larvae are not included in the culture. However, in the cell
culture of Toxorhynchites amboinensis, it is said that the larvae should
be injured so as to provide a minimum opening from which cells just
come out, but they should not be injured so severely so as to keep their
active movement in the culture (Kuno, 1981).
