Anchorage-Dependent Cell Culture
For anchorage-dependent cells, a roller bottle culture is commonly used,
although this type of culture has the disadvantage of a low surface-tovolume ratio .
• • Materials
Equipment - Roller bottles (Fig. 1)
- Microcarrier (glass beads, collagen beads, or plastic beads)
- Packed-bed reactors
- Spinner culture bottles
Solutions - Culture medium
• • Procedure
Preparation 1. Sterilize the components of the culture system.
2. Assemble the culture system aseptically.
Culture of 1. Harvest the cells from a stock culture.
cells 2. Inoculate the tank with the harvested cells.
3. Leave the cells for appropriate time to allow them to attach to the substrate.
4. Run the culture system .
• • Results
Cells grow by forming a monolayer on the inner wall of the roller bottle.
The cells alternatively come in contact with air and medium. This promotes the uptake of oxygen and nutrients by the cells.
