Microscope Photography
Photography is necessary to record the morphology of cultured cells.
Usually, cultured cells are observed using a phase-contrast optical system.
Materials
PROTOCOL
- A microscope that is able to be connected either to a photographic Equipment
apparatus or an automatic camera
- Photographic apparatus, consisting of an intermediate lens, a focusing lens, an instrument for measuring light intensity, shutter, and camera box
- Camera (an automatic camera should be able to determine exposure
time, and may be used without the photographic apparatus if it can
be connected to the microscope)
- Film-developing tank
- Enlarger
- Trays
- Safety lamp
- Developer (ND-4 developer: 0.2 g p-methylaminophenol sulfate Solutions
(metol), 1.125 g hydro quinone, 25.0 g sodium sulfite (anhydrous),
0.625 g sodium borate, 0.2 g citric acid per 500 ml distilled water)
- Fixative (240 g sodium thiosulfate, 15 g sodium sulfite (anhydrous),
13.5 g glacial acetic acid, 7.5 g boric acid, 15 g aluminum potassium
sulfate per 1,000 ml distilled water)
- Driwel (Fuji Photo Film)
- Stopper (1.5% acetic acid)
- Film (Minicopy HR II (Fuji Photo Film), Ektagraphic He (Kodak), Miscellaneous
Technical Pan (Kodak), or similar film)
- Photographic paper (no. 2 or no. 3; e.g. Mitsubishi, Fuji Photo Film,
Kodak, Agfa)

The following is a method of making black and white photographic
prints.
