Chapter 24
PROTOCOL
Echinodermata
Subprotocol 1
Gonads of Starfish (Asteroidea)
Materials
- Dissecting tray
Equipment
- Scissors
- Forceps
- Pasteur pipettes
- Petri dish
- Culture media (sterilized seawater)
Solutions
- Adults of starfish Asterias amurensis, Asterina pectinifera, or Asterina Animals
gibbosa
Procedure
1. Dissect the animals and excise the gonads.
Dissection
2. Cut, 10-12-mm pieces from the gonads.
1. Place the gonad fragments in a petri dish containing 4 ml culture me- Culture
dium (cf. Chaet et aI., 1964).
set-up
2. Culture at 14°-18°C.
Results
According to Kanatani (1964) and Kanatani and Ohguri (1966), when
ovary fragments from A. amurensis or A. pectinifera were cultured, they
released eggs during the culture period following stimulation by some
substance contained in the nerve ring-radial nerve system. Mecklenburg
and Chaet (1964) also reported that, in organ cultures of ovaries from A.
forbesi, nerve extract and Ca 2 + are necessary to induce shedding. Delavault
and Brusle (1965) and Brusle (1968) cultured whole gonads ofhermaphrodite starfish Asterina gibbosa in a medium consisting of 16 parts of
1 % gelatin, 2 parts of 1 % glucose, 1 part of 50% egg albumin, and 1 part
of 0.5% yeas to late (all solutions were made with sterilized seawater), and
25,000 !U/lOO ml penicillin G. Female dominant gonads showed considerable extension of the vesiculate tissue, and young ovocytes appeared
