30°C. Therefore, an incubator in which the temperature can be set between 5° and 40°C will be useful. Control of illumination is not necessary. In many insects diapause is induced by photoperiodism, whereas
in cultures photoperiodism does not affect the growth rate of insect cells.
It is better if cultures are kept under constant darkness.
A CO 2 incubator (Fig. 6), which is often used for culturing mammalian cells, is generally not necessary for invertebrate cell culture. In mammalian cell culture, a mixture of air with 5% CO 2 and saturated with
water vapor is used to keep the pH of the culture medium constant. However, most invertebrate cells are known empirically to be insensitive to
changes in the pH of culture media. Therefore, a CO 2 incubator is not
usually required for invertebrate cell culture. Of course, a CO 2 incubator
can be used, but an improvement in the culture by the use of the CO 2
incubator cannot be expected.
