Sterilization 1. Surface sterilize the animals with 20% Wesco dyne for 10 min.
2. Rinse the animals with 70% ethanol.
3. Air-dry the animals in a laminar flow cabinet for 5 min.
Dissection 1. Split the dorsal seam toward the rostrum and pull each half of the
carapace free of the thorax.
2. Remove testicular tissues carefully using forceps, avoiding contact with
the gut and midgut gland and other sources of contamination (Fig. 11).
3. Transfer testicular tissues into a drop of medium in a sterile petri dish.
