THE EFFECTS OF ADMINISTRATION OF PIMOZIDE AND (D—Arg , Trp , Leu

(SGnRH—A) ON GONADOTROPIN (GtH) RELEASE AND OVULATION IN
FISH.

Dopamine has direct GtH release—inhibitory activity in
of the dopamine antagonist pimozide (Pim) greatly potentiates the
se—response
to LHRH analogues. Mature female goldfish held at 20°C were '
injected intraperitoneally with Pim, at a dose of 10 Ug/g,and sGnRH—A,
a
dose of 0.1 or 0.01 ug/g, at the same time, or at two separate times
Pim followed 3 hours later by the peptide; in another experiment thesej
dosages of Pim and sGnRH—A were given in the form of an elastomer
implantation. The results demonstrate that PIM potentiates the response
sGnRH—A at both 0.1 and 0.01 ug/g, whether given at the same time or
hours
apart. Pellet implantation had no effects on Gth levels. The
ovulated females in groups given Pim and sGnRH—A at 0.1 and 0.01 ug/g was
similar (87—100Z). There were no ovulations in groups implanted with pellets}
In conclusion, injection of Pim (10 pg/g) and sGnRH—A (0.1 or 0.01
is effective in inducing ovulation in mature female goldfish whether the
two
are
given at the same time or with a 3 hours interval, but not if implant
ted as
the elastomer pellets. There were no differences in the viability
fertilized eggs,

to
the eyed stage, between the group injected with
and sGnRH—A, and Spontaneous ovulâted females.
