3. Adhesion
The next stage requires some “adhesive” to connect nucleic acid. Two approaches used for this are DNA ligase, which uses the adhesive surface to connect
DNA, and RNA ligase, which connects RNA. Genes or portions of genes removed
through (2) in the former are connected by promoters or vectors (Fig. 4.12).
4. Transcription and Cloning
Also essential is manipulation that freely “clones” information possessed by the
nucleic acids: from DNA to DNA (DNA polymerase), from RNA to RNA (RNA
replicase), from DNA to RNA (RNA polymerase), and from RNA to DNA (reverse
transcriptase; Fig. 4.13). Reverse transcriptase in particular is essential when
forming complementary DNA (cDNA) with information transcribed from
