7.7 Description of Semi-batch Study with Municipal
Wastewater
The semi-batch study was conducted in the fixed-bed hybrid bioreactor, already
fabricated using the municipal wastewater. For this purpose, the reactor was fed
with varying strength of municipal wastewater under different initial MLSS concentrations. The performance of the reactor was monitored at certain intervals over a
specific batch period.
7.7.1 Objective and Scope
The experimental plan was to check the response of the fixed-bed hybrid bioreactor
under semi-batch mode of operation. The semi-batch study was basically conducted
with an objective for performance evaluation of the fixed-bed hybrid bioreactor as
well as for determining the kinetic coefficients from the experimental results, which
are required for finding out the outputs from the mathematical model.
Scope of the semi-batch study was to determine the COD as well as mixed liquor
suspended solid (MLSS) concentration at various batch intervals over a set of batch
runs. At the same time, the attached biomass density was also measured at respective
batch intervals.
7.7.2 Procedure of Semi-batch Study
Semi-batch study was conducted in the laboratory-scale hybrid fixed biofilm reactor
at an interval of 3 h over a total 9 h batch period. Different biomass concentration
(MLSS) was set out at the initial time of batch operation. During this study, the
effluent sample was collected at each batch interval and analyzed for MLSS as
well as COD (substrate) concentration. In addition, biomass sample was extracted
from the attached surface to measure the density of the attached microorganisms.
Biomass density in the attached surface can be determined by washing the biofilm
surface with 25 ml 0.1 (N) NaOH and thereafter heating the solution at 80 °C [1]
followed by spectrometric observation of protein as per Lowry’s method. The total
biomass in the biofilm bioreactor (Xattached) was determined from a calibration
curve correlating protein and attached biomass. The results of semi-batch study with
municipal wastewater are shown in Table 7.3.
