Fig. 5.6 A Cartoon representation showing the secondary structure of S362CMoBOD variant
and its attachment to the maleimide-modified surface. The location of the engineered Cys residue
on the surface is highlighted in blue. B, C Bioelectrocatalytic waves of O 2 reduction at B a
S362CMoBOD- and C wild type MoBOD-modified electrodes by covalent attachment through
maleimide on GC/MWCNT electrode and by drop-casting 3 µL of the enzyme solution on the
electrode. The black and green curves show DET-type bioelectrocatalysis in deoxygenated and
O 2 -saturated solutions, respectively. The blue and red curves show MET-type bioelectrocatalysis in
deoxygenated and O 2 -saturated solutions, respectively, containing 0.3 mM ABTS at the same electrodes. Reproduced from Ref. [46] copyright (2019) (with minor modification) from The American
Chemical Society
to contain a stretch of six consecutive His residues at the N- or C-terminus were
attached via their affinities to the Ni–NTA moiety. Successful construction of an
oriented monolayer of cyt c oxidase on an Au electrode surface was demonstrated,
and DET-type bioelectrocatalysis of cyt c oxidase was confirmed [48, 49].
