37. If pest populations continue to grow after the use of beneficial
insects, insecticides like Enstar
® II, Tempo
® SC ultra, and
Conserve
® SC can be used to control pests. Insecticides should
be applied as directed on the label. Local regulations may
require any pesticide application to be performed by a licensed
pesticide applicator.
38. Marathon
® 1G, a granular insecticide, can be used as preventive insecticide or as treatment after an infestation. It can also
be mixed in with the soil or included in sub-irrigation watering.
Marathon
® should be applied as directed by the label. Local
regulations may require any pesticide application to be performed by a licensed pesticide applicator.
39. Keep the growth area clean and regularly sweep the floors
and/or shelves to eliminate or reduce potential sources of
pest outbreaks. Mature and dry plants should be harvested
and old soil and nonviable dry plant debris should be discarded
immediately.
40. Cut four equally spaced holes at the point where the sleeve
meets the top of the pot. This will increase aeration and reduce
water condensation that may encourage mold growth. Fold
down the tops of the sleeves about 2 cm to ensure they stay
open and stable. If plants grow out above the sleeves and are at
high plant density, train the top of the plants back down into
the sleeve to avoid contamination.
41. A 425 μm mesh screen sieve should be used when threshing
Arabidopsis. For other Brassicaceae species, since there is a
large variation of seed size, choose a sieve that has mesh marginally larger than the seeds. A large number of mesh screen
sieves are available that range in size from 200 μm to 3.35 mm.
42. When removing isolation sleeves, hold the plant away from
your work area to avoid contaminating it with seeds or growing
media.
43. After initial sieving, the seeds are still likely to be mixed with
soil and plant residue. A combination of additional sieving,
gentle blowing, and visual inspection can be employed to
clean the seeds completely. An alternative for a small sample
of seeds is to clean by hand with the aid of a pointed tool on an
opaque glass plate illuminated from below.
44. The moisture content of Arabidopsis seeds stored in open
containers corresponds to the room humidity. Arabidopsis
seeds behave in a similar way to crop seeds with similar chemical composition [13, 14].
45. The type of test that is used to determine if the variances are
equal is based on the number of samples/data sets. If there are
only two data sets, perform an F-test. For more than two data
sets, perform a Bartlett’s test.
