One full test can be completed in 30 days. However, because pre-cultivation of
the microcosm is necessary, culturing should be repeated several times before the
final assessment, and it should be confirmed that the control system (in which no
analyte was added) formed a stable system (Fig. 5.1). Microcosm N-system with
DO electrode and DO fluctuating pattern in microcosm were shown in Fig. 5.1.
5.2 Setup of a DO Meter in the Microcosm N-System
Install the DO electrode at the 14th culture time (336 h) and begin the
measurements. Setup of the electrode follows the methods described in Sects.
4.2.1 and 4.2.2. Care should be taken when installing the electrode to prevent
disturbance of the distribution of organisms in the microcosm. After setup, it is not
necessary to operate it aseptically when installing an electrode. However, the
electrode should be sterilized with alcohol to avoid trouble with the electrode,
which may be installed after confirming by smell that the alcohol has dried
(Fig. 5.2).
