– The nitrogen content of Polypeptone is 14.5% (w/w), and the phosphorus content
is 1.0% (w/w), as measured by the Japanese Industrial Standard (JIS) K-0102
method.
3.3 Microorganisms in the Microcosm N-System
The Kurihara-type microcosm N-system is a gnotobiotic microcosm in which the
species composition is completely known; experimentally, this is superior to the
stress-selected type of microcosm, in which environmental water is added to seed the
nutrient medium and culture. The naturally derived type is one in which the culture is
derived from the environmental water sampled from a natural lake or marsh in
equilibrium with and representative of the system that is necessary for impact
assessment. The microcosm N-system is a multi-species culturing ecosystem
model and consists of two species of Chlorophyceae, Chlorella sp. and Scenedesmus
quadricauda, and one species of cyanobacteria, Tolypothrix sp., as producers; one
species of ciliate protozoan (Cyclidium glaucoma), two species of rotifers (Lecane
sp. and Philodina erythrophthalma), and one species of oligochaete (Aeolosoma
hemprichi) as consumers (predators); and four species of bacteria, Bacillus cereus,
Pseudomonas putida, Acinetobacter sp., and a coryneform bacterium, as decomposers. The biological characteristics of each microorganism are described in Sects.
3.3.1, 3.3.2, and 3.3.3 below (Tanaka 2002; Sudo and Inamori 1997; Shiga Scientific
Teaching Materials Research Committee 2005; Japan Sewage Works Association
2012; Holt 1994).
