Production of EPN in Solid
Medium
Materials Required
l
Dog food chunks: 100 g.
l
Agar: 1 g.
l
Distilled water: 500 ml.
Procedure
1. Pulverize the dog food chunks in a blender.
2. Mix 100 g of pulverized dog food with 500 ml of dissolved
1% agar.
3. Blend the dog food/agar medium further for 1 min to obtain a
uniform mixture.
4. Autoclave for 15 min at 120
C.
5. Cool down the dog food agar medium after autoclaving at
60
C.
6. Pour 25 ml medium into sterile plastic petri dishes and inoculate with nematodes (100–200 IJs) on to the surface of the
medium.
7. Seal the plates and incubate at temperature of 28
C for
20–30 days.
8. For trapping of the IJs, remove the petri dish cover and the
bottom containing the nematode infested medium is placed
