There have been many kinds of aptamers generated by SELEX, such as biologically active ingredients, food safety hazards, biomarkers of disease, and so on.
However, there still have some disadvantages in the general SELEX procedure,
among which, the tedious experimental operations, the blindness, and uncertainty
of SELEX are the main problems, and thus, different SELEX variants have been
developed to improve the selection efficiency of SELEX.
1.3 Different SELEX Variants for Selection Rationalization
1.3.1 Reducing Screening Rounds
1.3.1.1 CE-SELEX
During the process of general SELEX, there are nonspecific interactions between
the target and their complexes, and it needs many rounds of selection to obtain
the candidates of aptamers with good specificity. Therefore, the process of general
SELEX is tedious and time-consuming. Some techniques as exemplified by capillary
electrophoresis (CE)-SELEX have been developed to reduce the round of SELEX.
CE-SELEX is developed by combining CE technique with SELEX, which is an
effective method to reduce the selection rounds of SELEX. The separation of CE can
be directly carried out in solution without introducing other separation media, there
is no nonspecific effect of aptamer and stationary phase separation media, and CE
possesses high separation efficiency for the separation of target molecules, aptamers
and their complexes. At the same time, the separation process can be easily completed
