5. Sterile gauze pads should be on hand in case of the need to
remove any excess blood around the incision site; they are
generally not needed.
6. A clean sterile instrument pack for each animal is recommended. If this is not possible, then instruments should be
wiped clean of blood and tissues with sterile gauze, rinsed in
sterile saline, and sterilized for 15 s using a glass bead sterilizer
or soaked in an appropriate chemical disinfectant according to
the manufacturer’s recommendations. If a glass bead sterilizer
is used, remember to allow time for the instruments to cool
before reusing them. If using a disinfectant, always rinse instruments with sterile saline before using on the next animal. When
in use, the tips of instruments should be kept within the sterile
field. Avoid touching unprepared areas. If this happens, replace
with a new sterile instrument or re-sterilize using bead sterilizer
or disinfectant as described above.
7. Dialysis membrane tubing may be prepared several days in
advance and stored at 4
C. Any unused tubing stored
>2 months should be autoclaved to ensure sterility. For that,
transfer to a 1-L glass beaker, cover with heavy-duty aluminum
foil, and autoclave for a minimum of 15 min at 121
C or 3 min
at 131
C. Once the beaker is cool to the touch, place in BSC
and transfer to a clean sterile container as described in Subheading 3.1, step 12.
8. Be sure to select the appropriate size glove for each user. Gloves
that are too large for the user’s hands will make it more difficult
to tie the dialysis tubing. Gloves that are too small may be
challenging to put on or lead to cramping during the
procedure.
9. The inner paper wrapping for sterile surgical gloves may be
used as a clean sterile workspace.
10. DMCs should be filled and used the same day. A small amount
(~10 ml) of high-BSA EMJH medium can be added to 50-ml
conical tubes used to store filled DMCs to avoid drying.
11. Depilatory creams can irritate the skin and, therefore, are not
recommended for hair removal. Preparation of the surgical site
should take place at a location (bench or room) that is separate
from where the surgical procedure will be performed.
12. Avoid wetting large areas of fur with chlorhexidine/isopropanol swab stick (or alcohol) because of the potential to induce
hypothermia.
13. During the scrub, the process should begin along the incision
line and extend outward and never from outward (dirty)
toward the center (clean).
14. A fresh pair of gloves is used for each animal.
