5. Discard disposable components of the system, such as microclaves and vent filters, in biohazard waste. Tubing, reservoirs,
caps/hardware, and any other reusable pieces of equipment
should be bagged and autoclaved out of the BSL3. After autoclaving, these items are soaked in tap water, then washed thoroughly with tap water to purge any medium residue, flushed
with deionized water, and then dried prior to reuse.
3.13 Biosafety
Considerations
Please refer to Notes 18 through 21.
4 Notes
1. Detailed descriptions of the reservoir caps, including the
PK/PD (in vitro toxicity) headpiece HF system hardware can
be found on the FiberCell Systems website (www.
fibercellsystems.com).
2. This section of tubing must be long enough to reach the threestop pump tubing and also allow for the medium tank to be
moved into the BSC. Alternatively, tubing from the media tank
may be routed through one or more 3-way pinch valves before
connecting with the peristaltic pump (Fig. 5). This permits
closure of the media flow line while allowing a feed from
another source, such as a drug reservoir, to be opened. Switching is made using a programmable controller wired directly to
the pinch valves (Fig. 5). Make tubing-to-tubing connections
using either double-ended or Y-shaped barb connectors. For
connections to other components, use Luer fittings with barbs.
3. The CR serves as a mixing chamber for medium and compounds before the solution is forced through the HF cartridge
by way of a piston operated “duet” pump, a binary action
mechanism that can drive two HF cartridges simultaneously.
4. This is an important factor that establishes the overall volume
of the system and is, therefore, a critical value in PK calculations
(see Subheading 3.2).
5. The system may also be sterilized in several pieces; use foil to
protect barbed connection points, and then assemble the components aseptically in a BSC to preserve system sterility.
6. In practice, we first attach sterile disposable microclaves to each
port, providing a self-sealing point of attachment for the syringes during medium loading and sampling of bacteria direct
from the ECS. Instructional videos demonstrating these and
other HF procedures are available on the FiberCell Systems
website (www.fibercellsystems.com).
