3 FISH on Chromosome Preparations of Peripheral Blood
79
Fig. I. Results of FISH experiments performed on chromosome preparations of healthy probands. Images were captured on a Zeiss Axioplan microscope using the ISIS digital FISH imaging system (MetaSystems, Altlussheim, Germany) using an XC77 CCD camera with on-chip integration (Sony). A The results of a FISH experiment using a whole
chromosome painting probe for chromosome 12 (red) and ofan mdm2-specific probe (green) in 12q13-14 are shown.
In the lower part of A the inverted DAPI-picture, which corresponds to the GTG-banding pattern of the human
chromosomes, is shown and it can be easily recognized that it is really chromosome 12 which has been painted.
B The result of a multi-color FISH experiment is presented. Apart from all 24 different whole chromosome painting
(wcp) probes, partial chromosome painting (pcp) probes for the long arms of nearly all human chromosomes have
been applied. The wcp probes have been labeled as described in Senger et al. (1998) and the pcp probes have been
labeled with a sixth fluorochrome (diethylaminocumarine). In the left upper part of Bthe inverted DAPI picture of the
metaphase is shown. C A multi-color FISH result using 7 different centromeric DNA-probes labeled with three different fluorochromes is shown (see also Nietzel et ai. 2001 and Chapter 32)
