31 Multicolor FISH Techniques for the Detection ofInter- and Intrachromosomal Rearrangements

- If background is a problem on the slides, apply an RNase digestion also
in combination with other proteolytic pretreatment procedures.
- A high background signal can also be caused by large amounts of DNA
probes in the hybridization mixture; high stringency posthybridization
washes at higher temperatures or lower salt concentrations may also
help.
- If probe specificity is poor and cross-hybridizations on other chromosomes occur, an increasing amount of con DNA in the hybridization
mixture may improve your results.
- For each labeling batch of DNA probes, check different probe and
con concentrations in the hybridization mix. This is most important,
when using new DNA probes which have never been checked before.
