The procedure of interphase FISH on human sperm cells is outlined in
Fig. 2.
Materials
Slide preparation - Quadriperm trays (Invitro Systems & Services)
- Otto's solution:
- 100 ml Tris-HCI buffer 10 mmolll, pH 7.4: 1.21 g Tris-aminomethan
and 1 I aqua bidest., adjust with HCI to pH 7.4
- 220 mg KCI (30 mmol/l)
- 100 mg MgCh x 6H z O (5 mmolll)
- 46 mg dithioerythritol (3 mmolll)
- Fixative: Acetic acid/methanol 1:4 (e.g. 50 ml acetic acid plus 150 ml
methanol, prepare freshly)
