Phylogeny of the Channichthyidae (Notothenioidei,
Teleostei) Based on Two Mitochondrial Genes

Introduction
The family Channichthyidae contains 11 genera and 15 species [1] and is
considered monophyletic [2,3-5]. Interrelationships of channichthyids were
first investigated by Iwami [2] based on morphological characters.
Some parts of I wami' s tree remained obscure, for instance the enigmatic
position of Dacodraco. The other study based on the osteological
development [6] failed to recover the same tree as in Iwami's. Therefore,
another marker which is independent from morphological characters is
necessary for this phylogenetic study. We explored the variability of four
different genes for several channichthyid species in order to investigate the
complete phylogenetic picture of the family. Among the genes studied,
three were mitochondrial: 16S mtDNA, cytochrome b, D-loop and one was
nuclear: 28S ribosomal DNA in its most variable domains D2 and D8.
To obtain the phylogeny of the family, we performed a total evidence
approach involving 1135 nucleotides from the the two informative genes
cytochrome b and the D-loop for 14 channichthyid species and one
outgroup.
Material and Methods
DNA sequences were obtained from the 11 channichthyid genera currently
recognized and 14 out of the 15 species of the family, plus one
Bathydraconidae as an outgroup. Details about their sampling are available
on request (Chen [7]). In order to examine intraspecific variation in the
most variable gene, the D-loop, three D-loop sequences from individuals of
Chionodraco hamatus from the population of Ross Sea, taken from
Bargelloni's unpublished data were used. To carry out the calibration of
molecular rate of the cytochrome b, available sequence of Trachurus
trachurus (Perciformes) was taken from GenBank under the accession
number ofX8l568.
