the continuous supply of culture medium, which prevents cell death due
to lack of nutrients, the production process is virtually independent of
the speed of infection (Jager, 1996).
Subprotocol 6
External Membrane Perfusion Culture
Almost any type of stirred tank or air-lift bioreactor can be combined
with external membrane perfusion. In the external membrane perfusion
system, the microfiltration module can be exchanged easily if the need
arises because of membrane blockage. However, the pumping of cells
and circulation through cross-flow microfiltration systems generate substantiallaminar shear stress to the cells .
• • Materials
Equipment - Culture system (any type of stirred tank or air-lift bioreactor)
- Perfusion module (hollow fiber, flat membrane, zigzag channel, and
spiral-like channel can be used)
Solutions - Culture medium
• • Procedure
1. Harvest cells from a stock culture.
2. Inoculate the tank with the cells.
3. Run the culture system.
