Equipment - Jar fermenter with a marine-type impeller
- Helical ribbon impeller (HRI) bioreactor
Solution - Culture medium
• • Procedure
1. Harvest cells from a stock culture.
2. Place the cells into the tank of the reactor.
3. Run the reactor .
• • Results
Koike and Sato (1988) cultured the Mamestra brassicae cell line NIASMaBr-93 in a 65-1 jar fermenter with a marine-type impeller. When the
cells were cultured in 40 1 MM medium containing 3% FBS, and an impeller rotation rate of 150 r. p.m., the cell density increased from 1.5 x 105
to 6.5 x lQ s cells/ml in 7 days. Kamen et al. (1991) cultured Sf-9 cells in
an 11-1 HRI bioreactor. This reactor was originally developed for highdensity plant cell suspension cultures. The structure of this tank is shown
in Fig. 4. The tank consists of a glass cylinder, top flange and sterile seal,
three surface baffles attached to the top flange and equally spaced, a profiled base, and a double HRI. The tank is equipped with a DO probe and
a pH probe to monitor the DO and pH of the medium, respectively, during operation. The impeller and surface baffling ensure homogeneous
mixing and high oxygen transfer through surface aeration and surfaceinduced bubble generation. In this culture system, cell density increased
