Preparation of Inoculum from an Infected Cell Culture
Once cultured cells are infected, the used culture medium contains extracellular viruses, and can be used as an inoculum.
Materials
Equipment - Centrifuge and centrifuge tubes
- Cell culture vessels
- Pasteur pipettes
Solution - Culture medium
Viruses - Virus inoculum
Procedure
Cell culture 1. Seed the cells.
2. Infect the cells with virus according to the qualitative inoculation
method (see subprotocol 5).
3. Once cultured cells are infected, the used culture medium contains
extracellular viruses, and can be used as an inoculum.
4. Collect the used culture medium.
S. Centrifuge the medium at ISO g for IS min.
6. Remove the supernatant and store it at -80 a C until use.
Results
The results are the same as those given for subprotocol 1.
Subprotocol 5
Qualitative Inoculation
In order to examine the sensitivity of the cells to certain viruses, or to
simply propagate a virus, cell cultures can be inoculated with an appropriate amount of the virus.
Materials
Equipment - Microscope
- Centrifuge and centrifuge tubes
- Culture vessels
- Pasteur pipettes
