If the cover glass is not clean, the cell suspension will not spread well. In
order to have a clean cover glass, treat the glass in the following manner:
- Wash the cover glass in neutral detergent
- Wash the cover glass under running water
- Keep the cover glass in 5% HCllonger than 1 day
- Rinse the cover glass under running water
- Immerse the glass in ethanol
- Wipe the glass with gauze or tissue paper
Results
Using the Giemsa stain, chromosomes are stained deep purple. If the
concentration of the hypotonic KCl solution used is appropriate, the chromosomes will disperse and will be able to be distinguished from each
other. If the concentration of KCl is too high, the cell membrane will
remain intact and the chromosomes will not disperse. If the concentration of KCl is too low, the cell membrane will burst during the hypotonic
treatment, and metaphase plates cannot be obtained.
