Culture 1. Transfer the organs into a culture vessel containing 0.25 ml culture
set-up
medium (see Fig. 2).
2. Press the organs gently against the surface of a cover slip placed in the
culture vessel.
3. Pool five culture vessels in a petri dish, keeping the atmosphere saturated with water from absorbent cotton soaked in sterile distilled water.
4. Maintain the petri dish in a humidified desiccator filled with an atmosphere of 95% air and 5% CO2 at 29°C .
• • Results
According to Seshan and Levi-Montalcini (197I), a large number of cells
migrated from explants of corpora cardiaca, whereas fewer cells migrated
from explants of corpora allata (Fig. 6). These cells were identified as
being mostly glial or interstitial cells. A few cells with neuronal characteristics were also seen in the migratory zone around explants of corpora cardiaca. The networks of cells migrating from corpora cardiaca
and corpora allata were maintained for 6-8 weeks.
