than half a light day. The bottles should be of the BOD type, made of white
borosilicate glass with ground glass stoppers. The kind of bottles and type of
glass of which they are made may influence upon rates of photosynthesis
(Worrest et al. 1980). Polycarboante plastic bottles of 200 ml can also be used
for measuring primary production in eutrophic waters and cold seas where the
bacterial overgrowth on their walls does not significantly influence the rate of
CO2 uptake. To decrease the microbial overgrowth on their walls, after the end
of the experiment they should be rinsed several times with hot (to - 60 0c) distilled water. Glass BOD bottles should be rinsed after use with a weak
acidified Lugol iodine solution prepared as follows: 2 g crystalline iodine is
mixed in the mortar with 4 g KI, and the mixture is gradually dissolved in
water. Its volume is then adjusted to 500ml, and 4ml of 1: 1 diluted HCI is
added. Some lOml of this solution is used to rinse the first bottle of the series.
Then this portion is transferred from bottle to bottle of the series after rinsing.
The bottles with the remaining Lugol solution are closed and kept until the
next experiment. This procedure prevents them from bacterial overgrowth on
