7.3 Description of Collection Point
Domestic wastewater from municipal wards of Kolkata Municipal Corporation
(KMC) is conveyed through a sewerage system and collected in various pumping
stations. In the present study, raw municipal wastewater was collected from a
pumping station of Kolkata Municipal Corporation located at Sealdah, West Bengal,
India. The wastewater was collected from underground pit, where the wastewater
from different places under respective sewerage zones is allowed to mix together.
The grab sample was collected from the pit manually to PVC jars as shown in Fig. 7.1.
7.4 Modality of Collection and Preservation Steps
Initially, municipal wastewater was collected periodically @ 30 L per day (carrying
each jar of 10 L) in two phases, Phase A and Phase B for characterization of raw
wastewater and biological acclimation with raw wastewater, respectively. Phase A
was continued from the summer period and Phase B was continued from the monsoon
period.
For the preservation of wastewater during characterization, biomass acclimation
and batch as well as continuous study, 1(one) ml of conc. H 2 SO4 was added per
liter of wastewater until pH value dropped below 2.0 and the same was kept in
the refrigerator. At the time of operation of the hybrid bioreactor with preservative
wastewater, necessary adjustment of pH up to about 7.2 was made with the help of
filtered lime solution.
