3.1.1 Preparation
of In-House Vaccine: In Situ
Adsorption of OVA onto
Alhydrogel
®
Alhydrogel
® is a commercially available 2% (w/v) aluminum
hydroxide wet gel. It is recommended to identify the optimal
binding efficiency between the antigen of interest and the aluminum (oxy)hydroxide particles in Alhydrogel
® to prepare a
vaccine [6].
1. Vortex Alhydrogel
® suspension briefly to break up the large
coagulates.
2. Use 5 N NaOH or 5 N acetic acid to adjust the pH to a desired
value, if needed.
