3. Other CAD software (e.g., Pro/Engineering, Siemens NX,
SolidWorks) as well as mesh modifying and slicing software
(e.g., MeshLab, Slic3r, Simplify3D) can be used.
4. The choice of the resin used in SLA can vary depending on the
need of the mechanical properties aimed, required compatibility with biological systems, and SLA printer used.
5. The degree of methacrylation on the gelatin can be modified to
alter the final mechanical properties of the hydrogel
[10]. Moreover, other liquid precursor solutions can be used
as coating material, such as alginate, collagen or collagen/
elastin, and neat thiol-ene solutions.
6. LAP can also be synthesized following the literature protocol
[11]. Other photoinitiators, such as 1-[4-(2-hydroxyethoxy)phenyl]-2-hydroxy-2-methyl-1-propanone (Irgacure 2959),
can be used to trigger the photopolymerization reaction in
combination with UV light.
7. The initial concentration of the hydrogel precursor solution
can be modified to alter the final mechanical properties of the
hydrogel.
8. Other options for UV light sources are available and depend on
the selected photoinitiator as well as the specific wavelengths
needed to initiate the photopolymerization reaction.
9. Collagen I-coated plates can be prepared by placing a 50 μg/
mL solution of collagen I, rat tail in 0.02 N acetic acid in
distilled water on standard tissue culture polystyrene plates
for 1 h at room temperature (e.g., ~7 mL for a standard T75
flask). Prior to use for cell culture, rinse with PBS.
10. Software available for segmenting digital images includes
Seg3D, 3D Slicer, ITK-SNAP, ImageJ, and InVesalius. Other
software such as MedCAD, Reverse engineering interface, and
STL interface can be used to convert medical images directly to
CAD models [8, 9].
11. Other software can be used for this purpose, see Note 3.
Moreover, other geometries can be fabricated, such as planar
mesh-like scaffolds on the mm to cm length-scales [7].
12. The current workflow can be adapted to other systems, such as
fused deposition modeling (FDM), melt electrospinning, and
selective laser melting (SLM). Based on the fabrication system
applied, different resolutions can be achieved.
13. Following treatment with the LIVE/DEAD
® kit, imaging
should be conducted within ~1 h.
