photo tube (Part No. 11551511), and a mechanical stage with
XY-drive (Part No. 11888705). A wide variety of microscope
brands and models can be used. Some minimum requirements
include the availability of stage clamps from Linkam Scientific
Instruments that are compatible with the microscope, the
availability of objectives with a free working distance of at
least 4.5 mm, and a condenser with a free working distance of
at least 12.7 mm (although Linkam Scientific Instruments also
makes available condenser extension lenses for many common
condenser models). The microscope must also include a camera
port and light source capable of generating ~3000 lumen (e.g.,
a 100-W, 12-V tungsten-halogen lamp).
6. The adapter should be selected to match the microscope camera port and the camera lens mount. Although C-mount hardware is available for Vision Research Phantom cameras, the
F-mount interface is preferable because it causes less
vignetting.
7. The temperature control system described in this chapter uses a
T95 system controller, a LNP95 liquid nitrogen pump, and the
Linksys32 software. The Linksys32 software was succeeded by
the LINK software in 2016, and the T95 controller was succeeded by the T96 model in 2017. New capabilities introduced
with the release of the LINK software greatly facilitate the
process of integrating a third-party high-speed video camera
and reduce synchronization error by providing improved data
sampling rates. Although specific instructions throughout this
chapter are applicable only to the model T95 controller hardware and the Linksys32 software interface, the basic experimental setup and workflow will remain the same if using the
T96 controller and LINK software.
8. For a high-speed video cryomicroscopy system based on the
model T95 controller and Linksys32 software, the BCS196
stage cable will not be used; instead, the BCS196 stage must
be connected to the system controller via a FDCS196 stage
cable, purchased separately.
9. Stage clamps should be selected to match the microscope.
Linkam Scientific Instruments Part No. 9670 is designed to
fit a Leica DM2500 M microscope with a mechanical XY-stage
(Part No. 11888705).
10. This component is required to establish communication
between the high-speed video camera and a Linkam cryomicroscope that uses a model T95 system controller and the
Linksys32 software. Note that if controlling the cryomicroscope system using the LINK software, then this item is not
needed; instead, synchronization with the camera can be
achieved using the logic signal input port on the Linkam
