1. Set the directory where the files are going to be saved.
2. Take the AFM head out, place the slide with the section on the
microscope stage, and stabilize the slide with magnetic clamps
(see Note 11).
3. Using an objective camera, move the sample to the center and
focus on the ROI in the sample (see Note 12).
4. Place the AFM head back on the microscope stage over the
sample (Fig. 3e).
5. In the Navigate window, adjust the focus on the cantilever by
moving the scan head down.
6. Using the objective camera, monitor the distance between the
tip and the sample. When the cantilever is close to the sample, it
should appear in the center of the view (Fig. 3f).
