annual species that may require vernalization to promote flowering.
The vernalization time varies depending on the species. For example, Thlaspi arvense can be induced to flower with a vernalization
period of 4
C for 21 days [8].
3.2.1 Maintenance
of Plants in Growth Area
Poor environmental conditions in a growth area, water stress, and
pest infestations can have a negative effect on plant health and seed
viability. Successful plant growth, silique/silicle production, and
good seed viability can be achieved when following a regimented
maintenance routine. This includes planting in a clean growth area,
a proper watering protocol, and proper pest management. The
following growth practices are useful for handling plants in any
growth context (greenhouse, growth chamber, or growth room).
The methods described have been designed for use with Arabidopsis but can be generalized to other species.
1. Remove and properly discard all plants and other materials in
the growth space.
2. Sweep and hose down the entire room interior (benches,
floors, window ledges, and windows) in a greenhouse or
sweep and clean all the surfaces with a disinfectant (e.g.,
Lysol
® ), then rinse with water in growth rooms or chambers.
3. Increase the temperature in the room to 40
C for 3–5 days (see
Note 29).
4. Do not place diseased or older plants in the clean room after
the high temperature treatment as to not introduce pest or
pathogens to young plants.
5. Move pot(s) to the growth area after planting (see Note 30),
add 2 cm of water to the tray (see Note 31).
6. Check the plants daily for water (see Notes 32 and 33).
7. Inspect sticky cards and plants daily for pests (see Notes 34 and
35).
8. Add appropriate beneficial insects as needed (see Note 36).
9. Apply pesticide if necessary (see Notes 37 and 38).
10. Maintain a clean growth area during plant growth (see Note
39).
3.2.2 Monitoring
Environmental Growth
Conditions
The environmental control systems currently offered for greenhouses, growth chambers, and growth rooms allow for remote
monitoring, control adjustment, and alarm notification via internet
connections. These features are a vital tool for avoiding loss of data
during plant production and maintaining control of environmental
conditions during experiments. Installation of remote sensing is
recommended for new growth facilities of all types.
