2. Keep the plate in the dark for at least 20 min.
3. Measure at the PAM fluorometer the quantum yield of photosystem II (F v /F m , where F v is the difference between the maximal (F m ) and the basal (F 0 ) fluorescence of chlorophyll).
Figure 3 shows that photosynthetic cell cultures maintained in
a culture medium containing 0.5% sucrose, although growing a bit
less than those maintained in 1% sucrose (Fig. 1l), exhibit the
highest photosynthetic efficiency (see Note 25).
