Automatic or Manual Activation?
If you know you will run a video at the moment you get to its slide, make it run
automatically. On the other hand, if you need to control the exact moment to get it
started (for instance, if you also introduce other elements on the same slide), use
manual activation (clicking with the mouse).
The automatic activation has the advantage of not depending on the mouse, which
can prove tricky with a nervous speaker. You can also consider delayed automatic activation, in which case, a bit of practice can help determining what would be the best delay.
Also, if your movie is short, consider having it run indefinitely in a loop, which
can help when a single viewing is not enough. Be careful though: if you talk while
you show the video, the audience will focus on the images, more than on your voice.
Unless what you say is directly related to what is going on in real-time on the video,
chances are they will stop listening to you.
