4.1.1 Handling
Nematodes
Materials Required

Nematode culture.
l
Glass pipette/suction device.
l
Dissecting needle.
l
Nylon bristle/fine wire/small wire loop.
l
Stereoscopic microscope.
l
Small plastic petri dish/watch glass.
Procedure
l
Take a dissecting needle to the end of which is attached with glue
a nylon toothbrush bristle, fine wire, or small wire loop to
handle single nematode in the suspension.
l
Let the nematodes be in shallow water, near the center of the
dish, and use microscope with suitable magnification to provide
the depth of focus and working distance.
l
Use the handling needle while viewing with the stereoscopic
microscope, to lift the nematode to the surface of the water;
then hold the bristle immediately beneath the nematode and
quickly flick up so that the nematode is drawn out over the
meniscus.
