the natural distribution of Nothofagus species in Argentina and/or natural forests
assays along environmental gradients (Arias Rios (N. alpina), Cagnacci (N. pumilio
and N. obliqua) and Sekely (N. pumilio) PhD theses).
Furthermore, the use of massive genotyping for population genomics studies in
Nothofagus species has been also validated, combining Ion Torrent PGM sequencing with the Fluidigm platform (48 individuals × 48 amplicons) as a cost-effective
strategy to produce re-sequencing data (El Mujtar et al. 2015). In total, ~50 barcoded libraries were sequenced on an Ion Torrent PGM across 4 runs, each being
either a single individual or different sizes (6 or 12 individuals) pools. Variation
between runs, techniques (either tagged individuals after mixing single PCR amplicons versus Fluidigm multiple PCRs) and single or pooled libraries for the same
technique and the impact of primer purification (desalting/PAGE) and of concentration of amplicons (equimolar/non-equimolar) were tested.
All these projects suggest that high-throughput sequencing and genotyping techniques could be more frequently used in Argentina for genome-wide association
studies (GWAS) and genomic selection (GS) in the near future accelerating forest
tree breeding of native Argentinean species.
